@Ari: You need to go online in Uplay and play the game online at least once. The game will make an update of somesort. Then you can keep Uplay and the game settings offline.
WARNING: If you play the game offline then you CAN NOT use the save when you log in online. The opposite is also true. If you have been playing it online and for some reason you can only play offline then you have to start from the beginning. Also, this post is misleading. You CAN NOT play the game without Uplay. You have to install Uplay and it will turn on whenever you start the game.
There will always be a background process running Uplay when you play the game. In fact you need BOTH Steam and Uplay running to launch the game. This post needs to be renamed as 'Playing offline with Uplay'.
